Intranasal PYY3-36 for ObesityDescription:PYY is a naturally occurring human hormone produced by specialized endocrine cells (L-cells) in the gut in proportion to the calorie content of a meal. Research has demonstrated the role of PYY in regulating appetite control. Therefore, PYY may represent a new approach to the treatment of obesity. Development Status:Nastech has recently initiated a dose ranging study designed to evaluate the pharmacokinetic parameters, as well as the effect on appetite, food intake and safety of various doses of PYY3-36 nasal spray in obese subjects. To date, Nastech has completed three Phase I trials of PYY. These trials have enrolled over 60 subjects and administered in excess of 900 doses in the Nastech PYY program. Results from the PYY Phase I clinical trials indicate the investigational product is safe, well-tolerated and shows evidence of reducing caloric intake, moderating appetite and demonstrating weight loss in human subjects. All PYY-related adverse events were mild or moderate in severity and resolved spontaneously without treatment. Clinical Need:Obesity is a chronic condition that affects millions of people and often requires long-term or invasive treatment to promote and sustain weight loss. In 2003, a study published in an issue of the Journal of the American Medical Association (JAMA) reported that the prevalence of obesity among U.S. adults was 20.9 percent in 2001. The latest estimates released by JAMA on April 5, 2006 shows that 32 percent of adults (over 66 million) and 17 percent of children and adolescents (2-19 years of age) are now obese. According to the U.S. Department of Health and Human Services, healthcare costs of American adults with obesity amount to more than $100 billion and approximately 300,000 U.S. deaths a year are associated with obesity and overweight.
